Subject: Starting new drug today AZD0530

Has anyone tried this new drug?? And if so what are the side effects you are having?? My husband has Thymoma cancer. It is recurring. This is a clinical trial, but the dr's seem very confident about this working. We feel that anything is better than doing nothing. Let me know if you have any information about this....

It looks like my partner might be starting this within the next couple of weeks.  She has SCC head/neck.  She finished radiation a month ago and the tumor has started to grow, so now they offered us a Phase II clinical trial with the AZD0530.  I have been trying to do some research on this.  In terms of side effects, I guess were going to see for ourselves soon, but our oncologist said that the side effects are comparatively minimal.  How is it going with your husband?

Hello,

  I must say the side affects were minimal. He had some moodiness, but the oncologist said that it wasnt from the meds. (I dont believe that). But we are no longer on this medicine as it didnt work for him. His tumors continued to grow. So he has been off this for about 3 weeks. Next week we are starting the other trial which is 5 days a week getting a drug intravenously and then 2 weeks off. and we continue this cycle for a while to see if it works.

We go to Sloan in NYC.  Pfister is our doctor.  I am hoping this will work.  We don't have a lot of options anymore. :(

Have you tried the other trial?? Its where you get 5 treatments in a row. They are given like chemo. And then you have 2 weeks off?? And do this all over again. The results are supposed to be a lot better than the one we were just on and you are about to try??

Our doctor mentioned another trial that we could do if this one didn't work.  I don't know the details of that trial, but it sounds as if those are the only two that she would be able to do.  Me, I'd prefer that they could go in there and cut the darn thing out.  Right now that doesn't seem to be a possibility.  We got the paperwork for the AZD0530 and I guess we'll sign off on it on Tuesday.  I guess we'll find out what the other one is if this one doesn't work....but maybe I can ask him on Tuesday.

I understand how it feels and all the unknowns you are facing right now. I dont evenknow what to say to you, other than I am there right now just like you. YOU need to keep your faith up and be supportive! I understand how you feel about wanting to go and just take the tumor out. But we did that and with type of cancer it does come back more times than not. I would be more than happy to keep in touch with you, I know I would have liked that if I had someone to do that with during this time. I have an email address I would have no problem giving to you. I have been on this for just over a month.  I have had some nausea from and some fatigue, but it isn't bad.  At the one month mark I had a CT to check for any possible lung damage (that is a possible side effect).  No growth, but it was just a month.  The more definative CT will be next month.  If there is no growth, or shrinkage, I will continue.

The other drug you mention is Belinostat.  I haven't heard or read that it is anymore effective or not.  Both AZD0530 and Belinostate were given Phase II status because there had been previous Phase I trials for solid tumors that included Thymoma patients that had positive results.  There is no indication that the results were statistically relavant or not

Also keep in mind that you don't have to confine yourself to trials specifically for Thymoma.  You can get into some Phase I trials for "solid tumors".  i was on one of those for 18 months starting in 2006.  There was an initial 20% shrinkage and then no growth for 15 months.  It was a Pfizer study with monoclonal anti-bodies IGFR-I receptor.  I understand that Mayo Clinic and IUPUI are currently negotiating with Pfizer for a Phase II trial specifically for Thymoma, but Pfizer does not want to finance the study.

There are also some other interesting Phas I trials that might come online soon.  One uses IGFR-I Monoclonal Antibodies in combination with other drugs like Cisplatin and Paclitaxel and another that uses some new molecular encapsulation of standard drugs that delivers the drugs only to targetted cancer cells with particular chemical receptors reducing or eliminating side effects allowing for higher doses or longer treatment of resistent tumors.

Hello,

    May I ask where and what type of cancer you have. Is it thymoma?? It sounds like you and my husband pretty much followed the same treatment plan. However you didnt mention a surgery. My husband did have the surgery. Im also interested in knowing if you had radiation??

   I was unaware of being able to be on other trials as you stated. I will be looking into that next. I always like to have another plan ready just in case.

Who is your Dr??
